If you are trying to connect to the mySQL database that is at your work and you have to use a VPN connection to remote into your desktop then you might have to VPN before you can connect to the mySQL server.

Yes, with backends you have to VPN onto the network if they behind a firewall and it depends on how the firewall is configured on what it is letting thru.
